Results: The researchers used genetic editing techniques to observe and compare the differences in bone trabecular volume and bone marrow fat content between these models and wild-type animals by constructing animal models with overexpression and knockout of the Ksr2 gene. Meanwhile, the researchers used molecular biology and imaging techniques to examine and analyze the bones of experimental animals in detail. This study demonstrates that the obesity gene Ksr2 has distinct effects on bone trabecular volume and bone marrow adiposity. Overexpression of the Ksr2 gene leads to a decrease in bone volume, whereas Ksr2 knockdown increases bone marrow adiposity. These findings provide insight into the complex relationship between obesity and bone health.

    KSR2 (Kinase suppressor of Ras2) protein is directly related to calcium store-manipulated calcium inward flow, and it plays an important role in the increase of cytoplasmic calcium concentration. KSR2 protein participates in the signaling pathway as a kinase component to enhance the cascade activation effect of the extracellular signal-regulated kinase (ERK) signaling pathway on calcium signaling. At the same time, the KSR2 protein connects and regulates the mitogen-activated protein kinase (MAPK) signaling pathway, an important energy receptor, which in turn interferes with and restricts the ERK signaling pathway. It has been shown that the KSR2 protein regulates the ERK signaling pathway and affects adipogenesis.
