Workflow

Sample Processing

First, we use lysis buffer or reagents to disrupt the cell membranes in the sample, releasing the cell contents. Cellular debris and other impurities are removed by centrifugation, filtration, or precipitation. Subsequently, we extract DNA from the sample using DNA extraction kits or chemicals. Common methods include using a mixture of phenol and chloroform to separate the DNA from other biomolecules or using magnetic beads to bind the DNA, which is then separated and eluted by a magnetic field.

Sample Preparation

We use restriction endonucleases to cut the DNA into fragments of a predetermined size and subsequently attach adaptors (junctions) to the ends of the DNA fragments. These junctions contain the primer binding sites and barcode information (for sample labeling) needed for sequencing. The DNA fragments with the junctions attached are subsequently amplified using polymerase chain reaction (PCR) to increase their concentration and add depth to the sequencing. The size distribution and quality of the sample are checked by gel electrophoresis or a bioanalyzer.

DNA Sequencing

The library is loaded into the chip or flow-through tank of the sequencing platform, and the sequencer is started for high-throughput sequencing. During the sequencing process, the instrument reads the sequence of the DNA fragments by synthesis or other techniques and generates raw sequencing data, such as FASTQ format files, which contain sequence information and quality scores for the sequenced reads.
