Targeted Bisulfite Sequencing
This method is used to assess site-specific DNA methylation changes. We use this method to obtain detailed information about the methylation status within the target region. The process involves DNA transformation by bisulfite, amplification using primers or probes, mixing of amplicons and junctions, and sequencing.
Reduced Representation Bisulfite Sequencing (RRBS)
RRBS performs enhanced sequencing based on bisulfite. It combines a combination of restriction endonuclease and bisulfite sequencing technologies to detect tens of thousands of unique CpG sites with broad coverage (CpG islands, genetic regulatory elements, gene promoters, repetitive DNA sequences, etc.). The process includes enzymatic digestion, library preparation, fragment screening, bisulfite transformation, PCR amplification, purification, and sequencing.
Whole Genome Bisulfite Sequencing (WGBS)
This method utilizes heavy sulfite treatment of unmethylated cytosine to convert it to uracil, which is amplified and converted to thymine, thus distinguishing it from methylated cytosine for detection purposes. We analyze the deep DNA methylation of the whole genome by WGBS, including methylation of CpG sites and methylation of non-CpG sites. This service includes raw data quality control, sequence alignment, methylation levels and analysis, sample correlation, clustering, PCA analysis, differential methylated region (DMR) screening, DMS/DMR related gene analysis, and DMR annotation.
Methylation-specific PCR
This is a common method for gene/sequence-specific detection of DNA methylation. We specifically detect methylation of genes to help analyze the role of DNA methylation in the regulation of gene expression and obesity. In the assay, we design specific primers to maximize amplification potential.
Global 5-mC Quantification
Accurate measurement of global 5-mC levels helps to understand the role of DNA methylation in biological processes. We analyze global 5-mC levels by enzyme-linked immunosorbent assay (ELISA) assay to quantify global DNA methylation in samples to understand the distribution of 5-mC.

Results: This study analyzed changes in gene expression and DNA methylation in visceral adipose tissue. The researchers analyzed gene expression differences, genome-level methylation, the effect of DNA methylation changes on gene expression, and the relationship with disease-related shapes in multiple subjects. A total of 11,120 differentially methylated CpGs (DMCs) and 96 DMRs were ultimately identified, with the MHC loci having the highest density of epigenomic variation. Sixty-eight genes in the obese group contained DMCs, and several genes were found to be significantly associated with glycosylated hemoglobin and fasting glucose. This study identifies new candidate genes associated with obesity and diabetes and demonstrates that DNA methylation can serve as a potential biomarker for obesity and diabetes.
