Summary: This study employed adenovirus-mediated overexpression of salt-inducible kinase 1 (SIK1) to investigate its role and mechanisms in hepatic glucose and lipid metabolism. Results indicate that SIK1 overexpression ameliorates hyperglycemia, hyperlipidemia, and fatty liver disease by downregulating phosphoenolpyruvate carboxylase (PEPCK), glucose-6-phosphatase (G6Pase), etc, while inhibiting gluconeogenesis and lipid deposition. This establishes SIK1 as a potential therapeutic target for type 2 diabetes and fatty liver disease.
