Handling Notes:

Summary:
The protein encoded by the LEP gene is secreted by white adipocytes into the systemic circulation, where it plays a pivotal role in the regulation of energy homeostasis. Circulating leptin acts by binding to its receptor in the brain, activating downstream signaling pathways that effectively inhibit feeding and promote energy expenditure. Beyond its central metabolic role, this protein possesses various endocrine functions and is involved in regulating processes such as immune and inflammatory responses, hematopoiesis, angiogenesis, reproduction, bone formation, and wound healing. Clinically, mutations in the LEP gene and its regulatory regions lead to severe obesity and morbid obesity with hypogonadism in human patients, and are further implicated in the development and progression of type 2 diabetes mellitus.
