JAK/STAT Pathway Functional Analysis Service

Inquiry

Overview

The JAK/STAT signaling pathway is essential for normal homeostasis and plays a key role in the signaling of a variety of cytokines and growth factors. Over 50 cytokines and growth factors, including interferons, hormones, interleukins, etc., have been discovered to be linked with the JAK/STAT signaling pathway. JAK/STAT also mediates apoptosis, inflammation, immune adaptation, tissue repair, and adipogenesis. When dysregulated, it leads to the development of obesity, diabetes, and other diseases. Protheragen offers a cutting-edge JAK/STAT pathway functional analysis service to better elucidate the mechanisms of obesity and to deeply analyze the Pathology of Obesity.

Unraveling Obesity Through JAK/STAT Pathway Insights

We explore the differences in the JAK/STAT pathway and its role in obesity in samples under different treatment conditions. In addition, we screen potential targets for this pathway and develop appropriate anti-obesity therapies. Combined with comprehensive preclinical analysis, we contribute to the research of innovative therapies.

JAK/STAT Pathway Functional Analysis

Our scope of analysis includes evaluating differences in JAK/STAT pathway activity in different samples, the effects of JAK/STAT pathway activation or inhibition on adipocyte function, metabolism, downstream molecules, etc. We are adept at mining deep information from obesity-related biomarker assays, protein expression, cell proliferation and apoptosis, and genes.

  • Sample processing
This process involves the cultivation and processing of animal and cell models, staining of tissue samples, RNA and protein extraction, etc. The entire sample processing process varies depending on the type of sample and specific experimental needs.
  • Pathway key factor analysis
This process includes a detailed analysis of JAK/STAT pathway-related genes and proteins using real-time PCR, enzyme-linked immunosorbent assay (ELISA), western blot, and other technologies.
  • Upstream and downstream signaling molecule analysis
We perform detailed detection and analysis of upstream and downstream signaling molecules of the JAK/STAT pathway. For example, we detect differences in various obesity-related biomarkers and cytokines (e.g., IL-6, TNF-α) by ELISA and other technologies.
  • Data analysis
Our team consists of experienced and highly specialized professionals covering a wide range of fields, including computational biology and bioinformatics. This ensures that we translate numerous experimental data into meaningful biological phenomena and explain in detail the correlation between JAK/STAT and obesity.

The indicators tested include, but are not limited to, the following components:

JAK1 STAT4 STAT1 STAT6 PIAS IFN-α/β
JAK2 STAT5a STAT2 TYK2 Shc IFN-γ
JAK3 STAT5b STAT3 IL-6 SHP IL-2
EGF GM-CSF GRB2 IL-12 IL-13 IL-3
IL-4 SOCS3

Anti-Obesity Therapy Development

Anti-Obesity Therapy Design

(Protheragen)

Potential therapies targeting the JAK-STAT pathway have been developed to combat diabetes and obesity. For example, a variety of small-molecule drugs and natural products have been found to modulate the JAK-STAT pathway for weight loss and lipid reduction. Based on our extensive obesity research, we offer the following anti-obesity therapeutic design and research services.

Preclinical Studies of Therapies

(Protheragen)

We provide one-stop preclinical research services for anti-obesity therapies. The scope of testing includes behavioral, body weight, leptin level, antioxidant capacity, oxidative stress, fat droplet accumulation, inflammation, obesity-related signaling pathway changes, insulin tolerance test, glucose tolerance test, etc.

Results: This study analyzed the use of cucurbitacin in the treatment of metabolic diseases. The results showed that cucurbitacin E (CuE) reduced adipogenesis and improved the expression of related genes in mouse adipocytes. It mediated the improvement of adipose tissue dysfunction and adipogenesis by attenuating JAK-STAT5 signaling and Jenus kinase signaling in adipose tissue. Cucurbitacin E can be used as a potential drug for the treatment of metabolic diseases.
