Targeting Sirtuins for Developing Anti-Obesity Therapeutics
InquiryOverview

Sirtuin (SIRT) is a nicotinamide adenine dinucleotide-dependent enzyme with a variety of biological activities. Several SIRTs are identified with different structures, cellular localisations, and functions. A growing number of studies have shown that SIRT is involved in different cellular metabolic pathways and plays an important role in the regulation of lipid metabolism, glucose, and adipocyte differentiation. This makes it an interesting therapeutic target for the treatment of obesity and its related complications. A number of potential anti-obesity drugs and therapeutic approaches have been screened and developed against SIRT. Based on in-depth research into the pathogenesis of obesity and cutting-edge assays, Protheragen offers a customized SIRT-targeting anti-obesity therapeutic development service. We provide flexible solutions around the anti-obesity therapeutic development process to support our clients throughout their research and help breakthroughs at every important point.
Your Best Partner for Study of SIRT-Targeting Anti-Obesity Therapies
After understanding the experimental needs of our clients, our talented and experienced team of scientists formulate innovative anti-obesity therapeutic development programs. We provide ongoing support for the entire therapeutic study, including design, screening, in vitro assays, in vivo studies, and safety assessments.
Anti-Obesity Therapy Development
Anti-obesity drug development: We are equipped with an experienced drug discovery team to provide one-stop service from anti-obesity drug design, screening, and synthesis to validation for our clients' research projects.
- Natural products: Studies have found that a variety of natural products have the potential to treat obesity and are a treasure trove of anti-obesity agents. For example, myricetin is a natural polyphenol that increases mitochondrial oxidative metabolism in adipose tissue by up-regulating SIRT expression, thus exerting anti-obesity effects. We have various diverse library of natural products with high throughput screening and experimental validation to obtain natural products with therapeutic potential for obesity.
- Other compounds: Given the role of SIRT in regulating glucose and lipid metabolism, appetite control, etc., SIRT modulators are potential drugs for the treatment of obesity and related complications. We design SIRT modulators by molecular modeling and analyze their binding affinity to their targets. Various compounds are then synthesized. Their cytotoxicity and anti-obesity effects are assessed by in vivo and in vitro experiments.
Other strategies: In addition to this, we are happy to develop other therapies targeting SIRT and evaluate their efficacy and safety in the treatment of obesity.
Preclinical Studies of Anti-Obesity Therapies
We are highly attentive to our client's needs and focus on the validation of a flexible approach to preclinical studies of anti-obesity therapies. Based on rigorous science, technology, and quality systems, each evaluation project maintains high quality and compliance.
Obesity models: We have a variety of obesity models, including in vivo (Mouse, Rat, Rabbit, Dog, etc.) and in vitro (adipocyte, etc.). These models are used to evaluate obesity mechanisms and related therapies. In addition, with years of experience and advanced technology, we also customize various obesity models according to the specific needs of our clients to maximize support for their research.
Evaluation of anti-obesity therapies: We provide one-stop preclinical research services for anti-obesity therapies, including pharmacodynamics, pharmacokinetics, bioanalysis, and safety assessment. We are proficient in data analysis and interpretation to extract meaningful insights from complex data sets.
Workflow
We carefully design each experimental session and provide comprehensive technical support to accelerate and advance the development and research of anti-obesity therapies targeting SIRT.
Applications
- Cancer research: SIRT modulators have an important role in the study of cancer treatment strategies in addition to being used in anti-obesity therapy research.

Results: This study analyzed the effects of myricetin on mitochondrial respiratory gene expression and lipolysis, as well as its potential in treating obesity. The researchers found that myricetin reduced triglyceride content in adipocytes and increased oxygen consumption rate and mitochondrial content. Also, myricetin feeding resulted in weight loss, improved glucose tolerance, and increased fatty acid consumption in mice induced by a high-fat diet. It increased Sirt3 expression and decreased mitochondrial protein acetylation. This study suggests that myricetin exerts its anti-obesity effects by up-regulating mitochondrial metabolism and Sirt3 expression.

Frequently Asked Questions
What are the types of sirtuin?
Seven types of SIRTs have been identified, with different structures and activities. SIRT1 is found in the nucleus and cytoplasm, has deacetylase activity, and regulates a variety of physiological processes, including cellular senescence and apoptosis, mitochondrial biosynthesis, and inflammatory responses. SIRT2 regulates gluconeogenesis, inflammation, and adipocyte differentiation through deacetylation. SIRT3 is located mainly in mitochondria and exerts metabolic regulatory effects by controlling mitochondrial biosynthesis and function. SIRT4 and 5 are mainly found in mitochondria. SIRT6 and 7 are mainly found in the nucleus. Deficiency of SIRT6 reduces lipolytic activity.
What are the advantages of natural products in anti-obesity drug development?
Natural products are widely available and structurally rich, making them an attractive source of anti-obesity agents. Many natural products alter metabolic processes and exert their anti-obesity effects by inhibiting pancreatic lipase and amylase, stimulating metabolism and thermogenesis, inhibiting adipogenesis, regulating appetite, enhancing insulin sensitivity, inducing apoptosis in adipocytes, and maintaining glucose homeostasis. Capsaicin, green tea, caffeine, and ephedrine have been suggested for obesity management.
