Summary: This paper focuses on the effects of interleukin-4Rα-deficient (IL-4Rα) in a high-fat, high-sugar diet-induced obesity model. IL-4Rα modulates high fat (HF) plus high carbohydrate (HC/fructose) diet (HF+HC) diet-induced weight gain, systemic adiposity, adipose tissue inflammatory gene expression, energy expenditure, exercise activity, glucose metabolism, hepatic steatosis, hepatic inflammatory gene expression, and hepatocyte injury. These effects were partially dependent on non-hematopoietic cell IL-4Rα expression but were not associated with direct signal transducer and activator of transcription 6 (STAT6) activation. Non-hematopoietic cell IL-4Rα was found to regulate the severity of high-fat, high-sugar diet-induced obesity and its associated sequelae through activation of nonclassical signaling pathways.
