Background: Triglycerides are major components of vegetable oils, animal fats, low-density lipoproteins (LDL), and very low-density lipoproteins (VLDL), and are carriers of fatty acids and sources of energy. Triglyceride turnover determines the utilization of fatty acids in mammalian tissues. Any dysfunction in this process may lead to changes in glucose metabolism, insulin resistance, and other diseases.

Assay Principle: Triglycerides are hydrolyzed by lipoprotein lipase to glycerol and free fatty acids. Glycerol is catalyzed by glycerol kinase (GK) to produce glycerol-3-phosphate and ADP, and glycerol-3-phosphate produces hydrogen peroxide by glycerol phosphate oxidase (GPO). In the presence of 4-aminoantipyrine and phenol, hydrogen peroxide is catalyzed by the enzyme peroxidase (POD) to produce quinone compounds, the content of which is proportional to the content of triglycerides.
