Description: Non-esterified free fatty acids (NEFAs) assay kit (Colorimetric, 0.01-3.0 mmol/L) is a metabolic assay kit for the rapid and sensitive detection of NEFA in the range of 0.01-3.0 mmol/L.
Assay Principle: The reaction of NEFA with coenzyme A, catalyzed by acetyl-CoA-synthetase (ACS), produces acetyl-CoA, which is catalyzed by acetyl-CoA oxidase (ACOD) to produce H2O2, which then reacts with TOOS and 4-amino-antipyrine (4-APP) to produce H2O2 under the catalytic action of peroxidase (POD) to produce a colored substrate. The colored substrate has a maximum absorption peak at 546 nm. The OD value at 546 nm is measured to calculate the NEFA content indirectly.
Background: Free fatty acids, also known as non-esterified fatty acids, are derived from the diet or metabolism of adipose tissue. In adipose tissue, hormone-sensitive lipase (HSL) breaks down triglycerides to produce glycerol and fatty acids. When circulating in the body, free fatty acids bind to plasma albumin and serve as a source of energy that is readily absorbed by muscle, brain, and other tissues and organs. Non-esterified free fatty acid (NEFA) is not only a product of fat hydrolysis, but also a substrate for fat synthesis. The concentration of NEFA has been linked to lipid metabolism, glucose metabolism, and endocrine function.
