Description:
Total cholesterol assay kit (Colorimetric) is a metabolic assay kit for the rapid and sensitive detection of total cholesterol in the range of 0.29-25.85 mmol/L.
Assay Principle:
Total cholesterol includes free cholesterol and cholesterol esters. Cholesterol ester can be hydrolyzed by cholesterol esterase to produce cholesterol and free fatty acids. Cholesterol ester is hydrolyzed by cholesterol esterase to produce cholesterol and free fatty acids. Cholesterol is oxidized by cholesterol oxidase to produce Δ4-cholestenone and hydrogen peroxide. In the presence of 4-aminoamylpyridine and phenol, hydrogen peroxide catalyzes peroxidase to produce a red quinone of benzoquinone imine phenizone, and the color shade of the resulting quinone is proportional to the cholesterol content. The cholesterol content of the sample is calculated by measuring the absorbance values of the standard and sample tubes separately.
Background:
Cholesterol is a sterol, a lipid found in cell membranes. Most cholesterol in the blood is in the form of cholesteryl esters. Lecithin-cholesterol acyltransferase in human plasma is an enzyme that catalyzes the formation of cholesteryl esters. Cholesterol synthesized or deposited in peripheral cells is returned to the liver via the reverse cholesterol transport system to reuse or recycle bile acids.
