Assay Principle:
Sucrase catalyzes the production of glucose from a substrate (sucrose), which, in the presence of glucose oxidase, produces hydrogen peroxide. Hydrogen peroxide reacts with the chromogen to produce a red substance with an intense absorption peak at 505 nm. Its absorbance is proportional to the concentration of glucose over a range of concentrations. Therefore, sucrase activity can be calculated by measuring the OD at 505 nm.
