Description: Low-density lipoprotein (LDL) cholesterol assay kit is a metabolic test kit for detecting uric acid by colorimetric method in the range of 0.2-12 mmol/L.
Background: LDL-cholesterol is the major lipoprotein in fasting plasma, accounting for approximately 2/3 of plasma lipoproteins, and is the main carrier of cholesterol to extrahepatic tissues. Defects in LDL-R function lead to a decrease in the clearance of plasma LDL-C, which ultimately leads to the formation of atherosclerotic plaques. Therefore, LDL-cholesterol levels are one of the main markers for disease detection.
Assay Type: Quantitative assay
Assay Principle: In the presence of surfactants, lipoproteins such as HDL and MHL (except LDL) change their structure and dissociate. The released micronized cholesterol molecules react with cholesterol enzyme reagents to produce hydrogen peroxide that is trapped in the absence of a coupling agent and is consumed without color development. At this point, the LDL particles remain intact. The addition of a reagent containing a coupling agent then dissociates the LDL particles, releasing cholesterol, which is catalyzed by cholesterol esterase (CE) and cholesterol oxidase (CO) to produce hydrogen peroxide. In the presence of 4-aminoantipyrine (4-AA) and phenol (T-OOS), hydrogen peroxide produces a red quinone compound catalyzed by oxidase (POD). The colored substance has a maximum absorption peak at 546 nm. The LDL cholesterol content of the sample can be calculated by measuring the OD value at 546 nm.
