Summary:
The PCSK1 gene produces a neuroendocrine protease that participates in the maturation of hormone and neuropeptide precursors along the secretory pathway. Initially synthesized as an inactive zymogen, the PCSK1 protein undergoes autocatalytic cleavage in the endoplasmic reticulum, followed by additional processing events in post-ER compartments that confer enzymatic activity. The mature protease accumulates in dense-core secretory granules and selectively cleaves substrates at basic amino acid motifs. PCSK1 activity is essential for proper endocrine signaling, and genetic disruption of this gene is linked to monogenic forms of obesity and proprotein convertase 1/3 deficiency. The gene generates multiple protein isoforms through alternative splicing.
