Summary: ENPP1 encodes an enzymatically active glycoprotein localized to the cell membrane as a type II transmembrane protein. Structurally, the functional unit consists of two identical subunits linked by disulfide bonds. ENPP1 exhibits enzymatic activity toward a wide range of nucleotide-based substrates, enabling cleavage of both phosphodiester and pyrophosphate linkages present in nucleotides and nucleotide sugars. Through these reactions, the protein contributes to nucleotide metabolism, including the conversion of nucleoside triphosphates into monophosphates and the degradation of diadenosine polyphosphates. Clinically, pathogenic ENPP1 variants have been reported in disorders involving ectopic calcification and skeletal abnormalities, as well as in metabolic conditions characterized by insulin resistance.
