Description:
Histone H3-K27 methylation assay kit is an epigenetic test kit that uses a colorimetric assay to measure histone H3-K27 methylation, which is fast and efficient and is completed in less than 3 hours.
Assay Principle:
In this assay, histones are stably spotted on bar wells. Methylated histone H3-K27 is recognized by high-affinity antibodies. The proportion or amount of methylated H3-K27 can be quantified by the HRP-conjugated secondary antibody chromogenic system and is proportional to the chromogenic intensity.
Background:
A major mechanism for the epigenetic inactivation of genes is the methylation of CpG islands in genomic DNA by DNA methyltransferases. Histone methyltransferases (HMTs) control or regulate DNA methylation through chromatin-dependent transcriptional repression or activation. G9a and polycomb group enzymes are HMTs that catalyze the methylation of histone H3 at lysine 27 (H3-K27) in mammalian cells. H3-K27 methylation participates in the silencing of gene expression at nonchromatin loci and, through the formation of polycomb binding sites, mediates heterochromatin formation. Mediate the formation of heterochromatin. Increased overall H3-K27 methylation has also been found to be associated with several pathological processes. Measurement of histone H3-K27 methylation is necessary.
