Description: Palmitic acid-13C16 is a fully carbon-labeled stable isotope optimized for use as an analytical internal standard for the quantification of native palmitic acid by GC- or LC-MS. As a ubiquitous 16-carbon saturated fatty acid accounting for 10-20% of human dietary fat intake, palmitic acid constitutes approximately 25% of human total plasma lipids and 65% of total saturated fatty acids, facilitating protein anchoring to the lipid bilayer and intracellular protein trafficking via protein acylation while regulating G protein-coupled receptor functions. Extensive cellular and in vivo data demonstrate that 200 µM of palmitic acid promotes migration and increases NF-kappaB p65 levels, MMP-9 activity, and ROS production in AsPC-1 pancreatic cancer cells, elevates COX-2 expression in RAW 264.7 cells, and enhances LPS-induced IL-1beta levels and caspase-1 activity in isolated mouse peritoneal macrophages; furthermore, a dietary regimen of 2.2% w/w palmitic acid for 12 weeks increases mouse hippocampal BACE1 activity and amyloid beta (1-42) levels, induces systolic contractile dysfunction in isolated mouse hearts, and shows marked elevation in the red blood cells of metabolic syndrome patients and the plasma of type 2 diabetes cohorts.
