Description:
Mouse agouti-related protein (AGRP) ELISA kit (31.2-2000 pg/mL) is a ready-to-use ELISA kit for analyzing mouse AGRP levels with a detection range of 31.2-2000 pg/mL and a sensitivity of 11.7 pg/mL.
Assay Principle:
The detection principle used in this kit is a sandwich enzyme immunoassay. The microtiter plate provided in the kit is pre-coated with an antibody specific for AGRP. Standards or samples are then added to the wells of the microtiter plate along with a biotin-conjugated antibody specific to AGRP. Avidin conjugated to HRP is then added to each microtiter plate well and incubated. Only wells containing AGRP, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change upon addition of TMB substrate solution. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is then measured spectrophotometrically at 450 nm ± 10 nm. The concentration of AGRP in the sample can then be determined by comparing the OD of the sample to a standard curve.
Background:
AGRP plays a role in body weight homeostasis, is involved in the control of eating behavior through the central melanocortin system, and acts as an inverse agonist of MC3R and MC4R, thereby inhibiting their constitutive activity. Genetic variants in AGRP may be responsible for obesity.
