Summary: MAGEL2 is a paternally expressed, imprinted gene located within the Prader–Willi syndrome (PWS) critical region on chromosome 15q11–q13. Loss of expression of genes in this region leads to PWS, a disorder characterized by neonatal hypotonia, developmental delay, hyperphagia, and childhood-onset obesity. MAGEL2 shares structural similarity with the neuronal differentiation factor necdin (NDN) and is thought to contribute to hypothalamic dysfunction underlying the obesity phenotype observed in PWS.
