Description: Human melanocortin 4 receptor (MC4R) ELISA kit (0.313-20 ng/mL, 0.188 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of MC4R in human samples with a range of 0.313-20 ng/mL and a minimum detectable dose (sensitivity) of 0.188 ng/mL. The kit is highly sensitive and easy to use.

Assay Principle: This kit is based on sandwich ELISA technology. An anti-human MC4R antibody is pre-coated onto 96-well plates, and a biotin-conjugated anti-human MC4R antibody serves as the detection antibody. Standards, test samples, and the biotinylated detection antibody are added to the wells sequentially, followed by a wash step with wash buffer. HRP-streptavidin is then added, and any unbound conjugates are cleared away with the wash buffer. TMB substrates are employed to visualize the HRP enzymatic reaction; the TMB is catalyzed by HRP to produce a blue product that changes to yellow after the addition of an acidic stop solution. The density of the yellow color is directly proportional to the amount of human MC4R captured in the plate. The O.D. absorbance is read at 450 nm in a microplate reader, allowing for the calculation of the human MC4R concentration.

Background: MC4R regulates adipogenesis and energy homeostasis by activating multiple intracellular and/or synaptic signaling molecules, while also modulating neural circuits. Its association with obesity has been well established. Studies reveal that MC4R mutations result in partial or complete loss of function, primarily linked to severe obesity. It is considered a potential monogenic cause of human obesity and represents a key therapeutic target for the disease.
