Description:
Mouse uncoupling protein 3 (UCP3) ELISA kit-Quantitative competitive is an ELISA-based in vitro research tool designed specifically for the quantitative detection of UCP3 in mouse samples with a minimum detectable dose (sensitivity) of 1.0 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This UCP3 ELISA kit utilizes a competitive enzyme immunoassay format featuring a polyclonal anti-UCP3 antibody and a UCP3-HRP conjugate. The test sample and buffer are incubated with the HRP-labeled conjugate in a pre-sensitized plate for sixty minutes. Because the specific binding sites on the polyclonal antibody are finite, the UCP3 in the sample competes with the UCP3-HRP conjugate for these positions. After five wash cycles, an HRP substrate is added to develop a blue product that turns yellow after the reaction is halted with the stop solution. The signal intensity at 450 nm is inversely proportional to the UCP3 concentration, which is then interpolated from a standard curve relating O.D. to standard concentrations.

In obesity research, UCP3 serves as a key regulator of thermogenesis and systemic energy balance, exhibiting high expression primarily in skeletal muscle. Studies indicate that elevated UCP3 levels may contribute to increased basal metabolic rate, enhanced fatty acid oxidation, and resistance to weight gain, positioning it as a potential therapeutic target for obesity treatment and improved insulin sensitivity.
