Description: Human prostaglandin H 2 (PGH 2 ) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PGH 2 in human samples with a range of 0.156-10 ng/mL and a minimum detectable dose (sensitivity) of 0.05 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: This experiment utilizes the double-sandwich ELISA technique. The pre-coated antibody is a human PGH 2 monoclonal antibody, and the detection antibody is a biotin-labeled polyclonal antibody. Samples and the biotinylated antibody are added to the microplate wells and subsequently washed with PBS or TBS. Then, avidin-peroxidase conjugates are added to the wells. After the reactants are thoroughly washed out with PBS or TBS, the TMB substrate is used for coloration. TMB turns blue through peroxidase catalysis and finally shifts to yellow under acidic conditions. The color depth is positively correlated with the concentration of the testing factors in the samples.
Background: PGH 2 is an intermediate product formed from arachidonic acid under the action of cyclooxygenase (COX), serving as the direct precursor for all prostaglandins, thromboxanes, and prostacyclins. These lipid mediators possess potent signaling functions. PGH 2 and its downstream products are extensively involved in processes including adipose tissue inflammation, adipocyte differentiation, angiogenesis, and insulin resistance.
