LipoKnoxa™ Human HSD11B1 shRNA AAV Particle (Silencing) focuses on the inhibition of HSD11B1, the enzyme that regenerates active cortisol within metabolic tissues. Silencing this gene using our U6 or miR30 systems provides a unique method to study the role of local glucocorticoid excess in visceral obesity. A wide range of AAV serotypes is provided for optimal tissue tropism, supported by total quality control screening for functional titer, sequence integrity, and sterility.

Summary:
HSD11B1 is a microsomal enzyme that serves as a primary local regulator of the potent stress hormone, cortisol. In metabolic tissues (adipose, liver), the enzyme primarily acts as a reductase, catalyzing the conversion of the inactive metabolite cortisone back into the active cortisol. Overactivity of HSD11B1, resulting in excessive local cortisol regeneration, is a key molecular contributor to central obesity and subsequent metabolic syndrome. A specific common variant in this gene has been clearly associated with increased risk of obesity and insulin resistance in children. Deficiencies involving HSD11B1 and H6PD lead to cortisone reductase deficiency.
