Description:
Human phosphoenolpyruvate carboxykinase 1; soluble (PCK1) ELISA kit (7.8-500 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PCK1 in human samples with a range of 7.8-500 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate included in this kit is pre-functionalized with a capture antibody specific to the target antigen. Standards or samples are introduced into the designated wells along with a biotin-conjugated detection antibody preparation specific for the target. Subsequently, avidin-conjugated HRP is added to each microplate well and incubated to form an immune complex. Following the addition of TMB substrate solution, color development occurs exclusively in those wells containing the target-biotin-avidin sandwich. The enzymatic reaction is terminated by a sulphuric acid solution, and the absorbance is measured spectrophotometrically at 450 nm ± 2 nm. The concentration of the target antigen in the specimens is determined by mapping the sample O.D. against the established standard curve.
Background:
Phosphoenolpyruvate carboxykinase 1 (PCK1) is a cytosolic enzyme that catalyzes the rate-limiting step of gluconeogenesis, converting oxaloacetate to phosphoenolpyruvate, thereby regulating glucose production from lactate and other TCA cycle intermediates. PCK1 plays a central role in maintaining glucose homeostasis and coordinating carbohydrate and lipid metabolism. Dysregulation of PCK1 activity can impair energy balance and contribute to abnormal fat accumulation, linking it to obesity and metabolic disorders.
