Description:
Human NAD-dependent protein deacetylase sirtuin-3; mitochondrial (SIRT3) ELISA kit (0.312-20 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT3 in human samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of 0.124 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit has been pre-immobilized with an antibody specific to SIRT3. Standards or samples are introduced into the appropriate microtiter plate wells along with a biotin-conjugated detection antibody preparation specific for SIRT3. Subsequently, avidin-conjugated HRP is added to each microplate well and incubated. After the addition of TMB substrate solution, color development occurs exclusively in wells containing the SIRT3 sandwich complex (antigen, biotinylated antibody, and enzyme-conjugated avidin). The enzymatic reaction is terminated with a sulphuric acid solution, and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 10 nm. The concentration of SIRT3 in the samples is then determined by comparing the optical density (O.D.) of the samples to the standard curve.
Background:
Sirtuin 3 (SIRT3) is a soluble mitochondrial NAD+-dependent protein deacetylase that regulates the activity of key metabolic enzymes, including glutamate dehydrogenase and isocitrate dehydrogenase 2, thereby influencing the citric acid cycle, antioxidant regeneration, and cellular energy metabolism. SIRT3 is processed in the mitochondrial matrix to its active form and plays a central role in maintaining mitochondrial protein acetylation and metabolic homeostasis.
