Description: Human cocaine- and amphetamine-regulated transcript (CART) ELISA kit (78.125-5000 pg/mL, 46.875 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CART in human with a range of 78.125-5000 pg/mL and a specificity of 46.875 pg/mL.
Assay Principle: Anti-human CART antibody is pre-coated onto a 96-well plate. Biotin-conjugated anti-human CART antibody is then used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibody are then added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to observe the HRP enzymatic reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of yellow color is proportional to the amount of human CART sample captured in the plate. The concentration of human CART can be calculated by reading the absorbance at 450nm with a microplate reader.
Background: CART is processed by prohormone/proprotein convertases to produce smaller, biologically active peptides and is considered to be involved in the regulation of appetite and stress. Its function is closely related to the actions of Leptin and Neuropeptide Y. Mutations in the gene are associated with obesity susceptibility.
