Description:
Rat plasminogen activator inhibitor-1 (PAI-1) ELISA kit (0.313-20 ng/mL, 0.188 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PAI-1 in rat samples with a range of 0.313-20 ng/mL and a minimum detectable dose (sensitivity) of 0.188 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This assay is based on sandwich enzyme-linked immunosorbent assay technology. A capture antibody is pre-immobilized onto 96-well plates, and a biotin-conjugated antibody is utilized for detection. Standards, test samples, and the biotinylated detection antibody are added to the wells sequentially, followed by a thorough wash with buffer. HRP-streptavidin is then introduced, and any unbound conjugates are cleared via a subsequent wash cycle. TMB substrates are employed to visualize the HRP-catalyzed enzymatic reaction, producing a blue product that turns yellow after the addition of an acidic stop solution. The intensity of the yellow signal is proportional to the amount of target analyte captured in the plate. The absorbance is read at 450 nm using a microplate reader, and the target concentration is calculated accordingly.
Background:
As the primary physiological inhibitor of tissue-type plasminogen activator and urokinase-type plasminogen activator, PAI-1 exerts its effects by suppressing fibrinolysis and proteolysis. Its elevation is associated not only with thrombosis and fibrosis but also with obesity and insulin resistance.
