Description: Rat 11 beta hydroxysteroid dehydrogenase type 1 (11-beta-HSD1) ELISA kit (1.563-100 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of 11-beta-HSD1 in rat samples with a range of 1.563-100 ng/mL and a minimum detectable dose (sensitivity) of 0.938 ng/mL. The kit is highly sensitive and easy to use.

Assay Principle: This kit is based on sandwich ELISA technology. An anti-rat 11-beta-HSD1 antibody is pre-coated onto 96-well plates. Standards, samples, and a biotin-conjugated detection antibody are added sequentially. After washing, HRP-streptavidin is introduced. TMB substrate is used to visualize the enzymatic reaction; the resulting blue color shifts to yellow upon adding an acidic stop solution. The yellow intensity is directly proportional to the amount of rat 11-beta-HSD1 captured. The O.D. is measured at 450 nm to calculate the final concentration.

Background: 11β-HSD1 is a key redox enzyme localized to the endoplasmic reticulum. It reversibly catalyzes the conversion of cortisol to the inactive metabolite cortisone. Although this enzyme exhibits bidirectional activity in vitro experiments, within living cells, it primarily converts inactive cortisone to active cortisol, thereby increasing local glucocorticoid concentrations. Obesity and type 2 diabetes are associated with abnormalities in glucocorticoid metabolism. The link between 11β-HSD1 and obesity represents a critical and extensively studied area of research.
