Description:
Rat NAD-dependent protein deacetylase sirtuin-3; mitochondrial (SIRT3) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT3 in rat samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of 0.088 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microtiter plate provided in this kit is pre-immobilized with a capture antibody specific to SIRT3. Standards or samples are added to the appropriate wells with a biotin-conjugated antibody preparation specific for SIRT3. Next, avidin-conjugated HRP is added to each well and incubated. Upon the addition of TMB substrate solution, a colorimetric change will occur exclusively in wells containing the SIRT3, biotin-conjugated antibody, and enzyme-conjugated avidin complex. A sulphuric acid solution halts the reaction, and the color change is measured at 450 nm ± 10 nm. The final concentration of SIRT3 is calculated by comparing the O.D. of the samples to the standard curve.
Background:
Sirtuin 3 (SIRT3) is a soluble mitochondrial NAD+-dependent protein deacetylase that regulates the activity of key metabolic enzymes, including glutamate dehydrogenase and isocitrate dehydrogenase 2, thereby influencing the citric acid cycle, antioxidant regeneration, and cellular energy metabolism. SIRT3 is processed in the mitochondrial matrix to its active form and plays a central role in maintaining mitochondrial protein acetylation and metabolic homeostasis.
