Description: Rat cocaine- and amphetamine-regulated transcript (CART) ELISA kit (15.62-1000 pg/mL, 5.9 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CART in rat with a range of 15.62-1000 pg/mL and a specificity of 5.9 pg/mL.
Assay Principle: The microtiter plate provided in this kit is pre-coated with an antibody specific for CART. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated antibody specific for CART. Avidin conjugated to horseradish peroxidase (HRP) is then added to each microplate well and incubated. Upon addition of the TMB substrate solution, only the wells containing CART, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is then measured spectrophotometrically at 450 nm ± 10 nm. The concentration of CART in the sample is then determined by comparing the OD of the sample to a standard curve.
Background: CART is processed by prohormone/proprotein convertases to produce smaller, biologically active peptides and is considered to be involved in the regulation of appetite and stress. Its function is closely related to the actions of Leptin and Neuropeptide Y. Mutations in the gene are associated with obesity susceptibility.
