Description: Rat ATP-citrate synthase (ACLY) ELISA kit (0.625-40 ng/mL, 0.375 ng/mL) is a ready-to-use ELISA kit for analyzing rat ACLY levels with a detection range of 0.625-40 ng/mL and a sensitivity of 0.375 ng/mL.
Assay Principle: The kit is based on the sandwich ELISA technique. Anti-rat ACLY antibody is pre-coated on a 96-well plate. Biotin-conjugated anti-rat ACLY antibody is used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibody are subsequently added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to observe the HRP enzymatic reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of sample captured in the plate. The concentration of rat ACLY can be calculated by reading the absorbance at 450 nm with a microplate reader.
Background: ACLY is the major enzyme responsible for the synthesis of cytoplasmic acetyl-CoA in many tissues. The product it catalyzes, acetyl-CoA, is used in several important biosynthetic pathways, including cholesterogenesis and lipogenesis.
