Product Overview

Description: Rat agouti-related protein (AGRP) ELISA kit (9.4-600 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of AGRP in rat with a range of 9.4-600 pg/mL and a specificity of 4.1 pg/mL.
Assay Principle: The microtiter plate provided in this kit is pre-coated with an antibody specific for AGRP. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated antibody specific for AGRP. Avidin conjugated to horseradish peroxidase (HRP) is then added to each microtiter plate well and incubated. Upon addition of TMB substrate solution, only the wells containing AGRP, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is then measured spectrophotometrically at 450 nm ± 10 nm. The concentration of AGRP in the sample can then be determined by comparing the OD of the sample to a standard curve.
Background: AGRP plays a role in body weight homeostasis. It participates in the control of feeding behavior via the central melanocortin system and is associated with obesity. By inhibiting the cAMP production mediated by the stimulation of melanocortin receptors in the hypothalamus and adrenal gland, it acts as an α-MSH antagonist.
