Description: Mouse proinsulin (PI) ELISA kit (0.156-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PI in mouse samples with a range of 0.156-10 ng/mL and a minimum detectable dose (sensitivity) of 0.094 ng/mL. The kit is highly sensitive and easy to use.

Assay Principle: An anti-mouse PI antibody is pre-coated onto 96-well plates. A biotin-conjugated anti-mouse PI antibody serves as the detection agent. Standards, samples, and the biotinylated antibody are added sequentially and washed. HRP-streptavidin is then introduced, and unbound conjugates are washed away. TMB substrate is used to visualize the HRP enzymatic reaction, which produces a blue product that shifts to yellow upon adding an acidic stop solution. The yellow signal density is directly proportional to the mouse PI concentration, measured at 450 nm.

Background: Proinsulin is the precursor molecule of insulin, synthesized in pancreatic β cells and subsequently processed into mature insulin and C-peptide. It plays a critical role in glucose homeostasis by serving as an essential intermediate in insulin biosynthesis and secretion. Under normal physiological conditions, proinsulin is efficiently converted to insulin; however, impaired processing or increased proinsulin secretion reflects β-cell dysfunction. Elevated circulating proinsulin levels are commonly observed in obesity and insulin resistance, making proinsulin a key indicator of metabolic stress and a contributing factor to the development of obesity-associated type 2 diabetes.
