Description:
Mouse phosphoenolpyruvate carboxykinase 1; soluble (PCK1) ELISA kit (1.56-100 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of PCK1 in mouse samples with a range of 1.56-100 pg/mL and a minimum detectable dose (sensitivity) of 0.39 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This assay employs a quantitative sandwich enzyme immunoassay technique. A capture antibody specific for PCK1 is pre-immobilized onto a microplate. Standards and samples are pipetted into the wells, allowing any PCK1 present to be bound by the stationary antibody. After the removal of unbound substances through washing, a biotin-conjugated detection antibody specific for PCK1 is introduced. Following a subsequent wash, avidin-conjugated HRP is added. After a final wash to remove any remaining unbound reagents, a substrate solution is added, and color develops in direct proportion to the amount of PCK1 captured in the initial step. The reaction is stopped, and the signal intensity is recorded to calculate the concentration.
Background:
Phosphoenolpyruvate carboxykinase 1 (PCK1) is a cytosolic enzyme that catalyzes the rate-limiting step of gluconeogenesis, converting oxaloacetate to phosphoenolpyruvate, thereby regulating glucose production from lactate and other TCA cycle intermediates. PCK1 plays a central role in maintaining glucose homeostasis and coordinating carbohydrate and lipid metabolism. Dysregulation of PCK1 activity can impair energy balance and contribute to abnormal fat accumulation, linking it to obesity and metabolic disorders.
