Description:
Mouse NAD-dependent protein deacetylase sirtuin-1 (SIRT1) ELISA kit (0.312-20 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT1 in mouse samples with a range of 0.312-20 ng/mL and a minimum detectable dose (sensitivity) of 0.117 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle:
The microplate included in this kit has been pre-immobilized with an antibody specific to SIRT1. Standards or samples are added to the appropriate microplate wells with a biotin-conjugated antibody specific to SIRT1. Subsequently, Avidin-conjugated HRP is added to each well and incubated. After the TMB substrate solution is introduced, only those wells containing SIRT1, the biotinylated antibody, and the enzyme-conjugated avidin will exhibit a color change. The enzyme-substrate reaction is halted by the addition of a sulphuric acid solution, and the absorbance is measured at 450 nm ± 10 nm. The SIRT1 concentration is then determined by mapping the O.D. values against the established standard curve.
Background:
Sirtuin 1 (SIRT1) is an NAD+-dependent protein deacetylase that links cellular energy status to transcriptional regulation, modulating processes such as metabolism, autophagy, apoptosis, and cell cycle control. SIRT1 deacetylates a variety of transcription factors and coregulators, influencing gene expression in liver, muscle, and adipose tissue, and participates in chromatin silencing through energy-dependent complexes. By sensing NAD+/NADH ratios, SIRT1 integrates metabolic signals and contributes to the regulation of glucose and lipid homeostasis, as well as cellular responses to stress and nutrient availability.
