Description:
Monkey brain cannabinoid receptor 1 (CB1) ELISA kit (0.5-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CB1 in monkey with a range of 0.5-10 ng/mL and a specificity of 0.1 ng/mL.
Assay Principle:
The microtiter plate is pre-coated with a monoclonal antibody specific to CB1. Standards or samples are then added to the wells of the microtiter plate, and if CB1 is present, it will bind to the wells pre-coated with the antibody. To quantify the amount of CB1 in the sample, a standard preparation of horseradish peroxidase (HRP)-conjugated CB1-specific polyclonal antibody is added to each well to “clamp” the CB1 immobilized on the microtiter plate. The microtiter plate is incubated, and the wells are then thoroughly washed to remove all unbound components. The substrate solution is then added to each well. The enzyme (HRP) and substrate will react during the short incubation period. Only wells containing CB1 and the enzyme-conjugated antibody will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm. A standard curve of color intensity (O.D.) versus standard concentration is plotted. The concentration of CB1 in each sample is derived from this standard curve.
Background:
CB1 is a member of the cannabinoid receptor family and the G protein-coupled receptor (GPCR) superfamily. It is expressed at the highest levels in different brain regions, and is also present at lower levels in most other cell/tissue types, including adipose tissue. It is thought to activate multiple signaling pathways, regulating energy metabolism and cell survival/death. CB1 has become a drug target for treating obesity. CB1 inhibitors may be potential anti-obesity drugs.
