Description: Human proinsulin ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of proinsulin in human samples with a minimum detectable dose (sensitivity) of <10 pg/mL. The kit is highly sensitive and easy to use.

Assay Principle: The human proinsulin ELISA kit is designed for the quantitative in vitro measurement of human proinsulin in cell culture supernatants. This assay utilizes a specific antibody pre-coated onto a 96-well plate. Standards and samples are pipetted into the wells, where any proinsulin present is captured by the immobilized antibody. Following a wash step, a biotinylated anti-human proinsulin antibody is added. After washing away the unbound biotinylated antibody, streptavidin-conjugated HRP is introduced. After a final wash, TMB substrate solution is added, resulting in color development in direct proportion to the amount of bound Proinsulin. The stop solution shifts the color from blue to yellow, and the intensity is measured at 450 nm.

Background: Proinsulin, the inactive precursor of insulin, is synthesized in pancreatic beta cells and enzymatically cleaved into active insulin and C-peptide. In early obesity, proinsulin cells undergo compensatory hyperplasia and secrete large amounts of insulin. However, as obesity worsens and metabolic stress increases, proinsulin-producing cells begin to fail, reducing the conversion efficiency of proinsulin to insulin. This leads to an elevated ratio of proinsulin to insulin in the bloodstream, serving as a key predictor for the future development of type 2 diabetes in obese individuals.
