Description: Canine brain cannabinoid receptor 1 (CB1) ELISA kit (0.5-10 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CB1 in canine with a range of 0.5-10 ng/mL and a specificity of 0.1 ng/mL.
Assay Principle: The assay sample and buffer are incubated together with CB1-HRP conjugate in a pre-coated plate for one hour. At the end of the incubation period, the wells are decanted and washed five times. The wells are then incubated with HRP enzyme substrate. The products of the enzyme-substrate reaction form a blue color complex. Finally, the reaction is stopped by the addition of a stop solution, which turns the solution yellow. The color intensity is measured spectrophotometrically at 450 nm on a microplate reader. The color intensity is inversely proportional to the concentration of CB1 because the CB1 in the sample competes with the CB1-HRP conjugate for the binding site of the anti-CB1 antibody. Since there is a finite number of binding sites, as more sites are occupied by CB1 in the sample, fewer sites remain to bind the CB1-HRP conjugate. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of CB1 in each sample is derived from this standard curve.
Background: CB1 is a member of the cannabinoid receptor family and the G protein-coupled receptor (GPCR) superfamily. It is expressed at the highest levels in different brain regions, and is also present at lower levels in most other cell/tissue types, including adipose tissue. It is thought to activate multiple signaling pathways, regulating energy metabolism and cell survival/death. CB1 has become a drug target for treating obesity. CB1 inhibitors may be potential anti-obesity drugs.
