Description: Human NAD-dependent protein deacetylase sirtuin-1 (SIRT1) ELISA kit (78-5000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT1 in human samples with a range of 78-5000 pg/mL and a minimum detectable dose (sensitivity) of 32 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle: The microtiter plate provided in this kit has been pre-immobilized with an antibody specific to the target antigen. Standards or samples are added to the designated microtiter wells along with a biotin-conjugated antibody preparation specific for the target antigen. Avidin-conjugated HRP is then added to each well and incubated. After the TMB substrate is added, only those wells containing the target antigen complex will exhibit a colorimetric change. The reaction is terminated by a sulphuric acid solution, and the color change is measured spectrophotometrically at 450 nm ± 2 nm. The concentration of the target antigen is determined by mapping the O.D. of the samples against the standard curve.

SIRT1 deacetylates a variety of transcription factors and coregulators, influencing gene expression in liver, muscle, and adipose tissue, and participates in chromatin silencing through energy-dependent complexes. By sensing NAD+/NADH ratios, SIRT1 integrates metabolic signals and contributes to the regulation of glucose and lipid homeostasis, as well as cellular responses to stress and nutrient availability.
