Description: Human NAD-dependent protein deacetylase sirtuin-1 (SIRT1) ELISA kit (0.781-50 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SIRT1 in human samples with a range of 0.781-50 ng/mL and a minimum detectable dose (sensitivity) of 0.28 ng/mL. The kit is highly sensitive and easy to use.

Assay Principle: The microtiter plate provided in this kit is pre-coated with an antibody specific to SIRT1. Standards or samples are introduced into the designated wells along with a biotin-conjugated detection antibody preparation specific for SIRT1. Next, avidin-conjugated HRP is added to each microplate well and incubated. Upon the addition of TMB substrate solution, color development occurs exclusively in those wells containing the SIRT1-biotin-avidin complex. The enzymatic reaction is terminated with a sulphuric acid solution, and the color change is measured spectrophotometrically at 450 nm ± 10 nm. The concentration of SIRT1 in the samples is determined by comparing the O.D. of the samples to the standard curve.

SIRT1 deacetylates a variety of transcription factors and coregulators, influencing gene expression in liver, muscle, and adipose tissue, and participates in chromatin silencing through energy-dependent complexes. By sensing NAD+/NADH ratios, SIRT1 integrates metabolic signals and contributes to the regulation of glucose and lipid homeostasis, as well as cellular responses to stress and nutrient availability.
