Description: Human carboxypeptidase E (CPE) ELISA kit (0.625-40 ng/mL, 0.221 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of CPE in human with a range of 0.625-40 ng/mL and a specificity of 0.221 ng/mL.
Assay Principle: The microtiter plate provided in this kit is pre-coated with an antibody specific for CPE. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated antibody preparation specific for CPE. Avidin conjugated to horseradish peroxidase (HRP) is then added to each microplate well and incubated. Upon addition of the TMB substrate solution, only the wells containing CPE, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm ± 10 nm. The OD of the sample is then compared to a standard curve to determine the concentration of CPE in the sample.
Background: CPE is a peripheral membrane protein that cleaves C-terminal amino acid residues and participates in the biosynthesis of peptide hormones and neurotransmitters. This protein specifically binds to proteins in the regulated secretory pathway, including prohormones, but does not bind to constitutively secreted proteins. Studies have found that mutations in the gene are related to obesity and Type 2 diabetes.
