Description:
Human agouti-related protein (AGRP) ELISA kit (<4 pg/mL) is a ready-to-use microwell, strip, or full plate ELISA kit for analyzing human AGRP levels with a sensitivity of <4 pg/mL.
Assay Principle:
The kit is based on a standard sandwich ELISA technique. Mouse monoclonal antibody specific for AGRP has been pre-coated on a 96-well plate. Standards and test samples are added to the wells, followed by biotinylated goat polyclonal antibody specific for AGRP, and washed with PBS or TBS buffer. Avidin-biotin-peroxidase complex is added, and unbound conjugate is washed with PBS or TBS buffer. The HRP substrate TMB is used to visualize the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of human AGRP sample captured in the plate.
Background:
AGRP plays a role in body weight homeostasis, is involved in the control of eating behavior through the central melanocortin system, and acts as an inverse agonist of MC3R and MC4R, thereby inhibiting their constitutive activity. Genetic variants in AGRP may be responsible for obesity.
