Description:
Human α2-antiplasmin (α2-AP) ELISA kit is an ELISA-based in vitro research tool designed specifically for the quantitative detection of α2-AP in human with a range of 0.5-10 ng/mL and a specificity of 0.1 ng/mL.
Assay Principle:
The microtiter plate is pre-coated with a monoclonal antibody specific to α2-AP. Standards or samples are then added to the wells of the microtiter plate, and if α2-AP is present, it will bind to the wells pre-coated with the antibody. To quantify the amount of α2-AP in the sample, a standard preparation of horseradish peroxidase (HRP) -α2-AP specific polyclonal antibody is added to each well to “clamp” the α2-AP immobilized on the microtiter plate. The microtiter plate is incubated, and the wells are then thoroughly washed to remove any unbound components. The substrate solution is then added to each well. The enzyme (HRP) and substrate will react during the short incubation period. Only wells containing α2-AP and enzyme-bound antibody will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is measured spectrophotometrically at 450 nm. A standard curve is plotted between color intensity (O.D.) and standard concentration. The concentration of α2-AP in each sample is extrapolated from this standard curve.
Background:
α2-AP is a plasma serpin (serine protease inhibitor) that is the principal inhibitor of plasmin, thereby playing a key role in stabilizing blood clots and preventing excessive fibrinolysis.
