Assay Principle: An antibody specific for 11β-HSD1 has been pre-coated on a microtiter plate. Standards and samples are pipetted into the wells, and any 11β-HSD1 will bind to the immobilized antibody. After removing unbound material, a biotin-conjugated antibody specific for 11β-HSD1 is added to the wells. After washing, streptavidin-conjugated HRP is added to the wells. After washing to remove unbound avidin-enzyme reagent, a substrate solution is added to the wells, and the color develops in proportion to the amount of 11β-HSD1 bound in the initial step. The color development is stopped, and the color intensity is measured.

Background: 11β-HSD1 catalyzes the reversible conversion of cortisol to the inactive metabolite cortisone and plays a pathogenic role in obesity and metabolic syndrome. It could be utilized as a potential target for anti-obesity studies.
