Description: Mouse enoyl coenzyme A hydratase, short chain, 1, mitochondrial (ECHS1) ELISA kit (78-5000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of ECHS1 in mouse with a range of 78-5000 pg/mL and a specificity of 39 pg/mL.
Assay Principle: The microtiter plate provided in this kit is pre-coated with an antibody specific to mitochondrial ECHS1. Standards or samples are then added to the appropriate microtiter plate wells along with a biotin-conjugated polyclonal antibody preparation specific for ECHS1, and avidin conjugated to horseradish peroxidase (HRP) is added to each microtiter plate well and incubated. TMB substrate solution is then added to each well. Only wells containing ECHS1, mitochondria, biotin-conjugated antibody, and enzyme-conjugated avidin will show a color change. The enzyme-substrate reaction is terminated by the addition of sulfuric acid solution, and the color change is then measured spectrophotometrically at 450 nm ± 2 nm. The OD of the sample is then compared to a standard curve to determine the concentration of mitochondrial ECHS1 in the sample.
Background: ECHS1 belongs to the enoyl-CoA hydratase/isomerase family. It functions in the second step of the mitochondrial fatty acid beta-oxidation pathway. It catalyzes the hydration of 2-trans-enoyl-coenzyme A (CoA) intermediates to L-3-hydroxyacyl-CoAs.
