Description: Rat tumor necrosis factor soluble receptor II (TNFSR-II) ELISA kit-Quantitative sandwich is an ELISA-based in vitro research tool designed specifically for the quantitative detection of TNFSR-II in rat samples with a range of 62.5-4000 pg/mL and a minimum detectable dose (sensitivity) of 15.6 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle: This assay employs a quantitative sandwich enzyme immunoassay technique. A capture antibody specific for TNFSR-II is pre-immobilized onto a microplate. Standards and samples are pipetted into the wells, allowing any TNFSR-II present to be bound by the stationary antibody. After the removal of unbound substances through washing, a biotin-conjugated detection antibody specific for TNFSR-II is added. Following a subsequent wash, avidin-conjugated HRP is introduced. After a final wash to clear any remaining unbound reagents, a substrate solution is added; color develops in proportion to the amount of TNFSR-II captured in the initial step. The reaction is then stopped, and the intensity of the color is measured to determine the concentration of the target analyte.
Background: TNFSR-II is the free form released into the bloodstream following proteolytic cleavage of the membrane-bound TNF-RII receptor (TNFRSF1B). TNFSR-II primarily functions as a decoy receptor, capable of binding circulating tumor necrosis factor TNF-α to neutralize its activity and reduce its binding to membrane-bound receptors, thereby negatively regulating systemic inflammation. In obesity research, serum TNFSR-II concentration is a recognized and reliable biomarker of chronic low-grade inflammation and endothelial dysfunction. Elevated TNFSR-II levels are significantly associated with central obesity, insulin resistance, metabolic syndrome, and subsequent cardiovascular disease risk. Consequently, it is frequently used to assess inflammatory burden and disease progression in obese patients.
