Description: Rat tartrate-resistant acid phosphatase 5b (TRACP 5b) ELISA kit (0.63-40 ng/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of TRACP 5b in rat samples with a range of 0.63-40 ng/mL and a minimum detectable dose (sensitivity) of 0.38 ng/mL. The kit is highly sensitive and easy to use.

Assay Principle: The micro ELISA plate is pre-coated with an antibody specific to rat TRACP-5b. Standards or samples are added to the wells to combine with the immobilized capture antibody. A biotinylated detection antibody specific for rat TRACP-5b and an avidin-HRP conjugate are then added successively and incubated. Following a wash to remove free components, a substrate solution is added. Wells containing the target will appear blue and then turn yellow after the stop solution is applied. The absorbance (OD) is measured at 450 ± 2 nm, and the OD value is proportional to the concentration of rat TRACP-5b.

Background: TRACP 5B, a metallophosphoesterase located at 19p13.2, encodes a tartrate-resistant acid phosphatase (EC 3.1.3.2) essential for bone resorption and the negative regulation of inflammatory cytokines, including TNF and interleukin-1 beta. By dephosphorylating osteopontin and bone sialoprotein, it maintains skeletal integrity and immune homeostasis; consequently, inactivating mutations lead to SPENCDI, while its elevated expression serves as a biomarker for various leukemias and Gaucher disease. In the context of metabolic health, TRACP 5B is increasingly recognized for its role in obesity-driven chronic inflammation, as its regulation of osteopontin signaling and macrophage activity directly influences the adipose tissue remodeling and systemic insulin resistance associated with excessive weight gain.
