Description: Rat syndecan-3 (SDC3) ELISA kit-Quantitative competitive is an ELISA-based in vitro research tool designed specifically for the quantitative detection of SDC3 in rat samples with a range of 2.5-50 ng/mL and a minimum detectable dose (sensitivity) of 0.1 ng/mL. The kit is highly sensitive and easy to use.
Assay Principle: The SDC3 ELISA kit utilizes a competitive enzyme immunoassay technique, employing a polyclonal anti-SDC3 antibody and an SDC3-HRP conjugate. Samples and assay buffers are co-incubated with the SDC3-HRP conjugate in a pre-coated plate for one hour. During this period, SDC3 from the samples and the SDC3-HRP conjugate compete for a limited number of anti-SDC3 antibody binding sites. Following incubation, the wells are decanted and washed five times. The wells are then incubated with an HRP substrate, forming a blue complex that turns yellow upon the addition of a stop solution. The intensity of the color, measured at 450 nm, is inversely proportional to the SDC3 concentration. A standard curve is plotted to relate the color intensity (O.D.) to the concentration of the standards, and the SDC3 concentration in each sample is interpolated from this curve.
Background: SDC3 is a major cell surface heparan sulfate proteoglycan belonging to the Syndecan family, playing crucial roles in cell adhesion, signal transduction, and cytoskeletal organization. The core function of SDC3 lies in its ability to bind to various growth factors and cytokines through its glycosaminoglycan chains, thereby regulating extracellular signaling. In obesity research, SDC3 is particularly critical as it is considered a pivotal regulator of appetite and energy balance. Highly expressed in the hypothalamus, SDC3 directly participates in controlling satiety and food intake. It represents a potential therapeutic target for obesity, especially for central nervous system-mediated appetite dysregulation.
