Description:
Rat neurotrophic tyrosine kinase receptor type 2 (NTRK2) ELISA kit (93.75-6000 pg/mL) is an ELISA-based in vitro research tool designed specifically for the quantitative detection of NTRK2 in rat samples with a range of 93.75-6000 pg/mL and a minimum detectable dose (sensitivity) of 56.25 pg/mL. The kit is highly sensitive and easy to use.
Assay Principle:
This kit is based on sandwich ELISA technology. An anti-rat NTRK2 antibody is pre-coated onto 96-well plates, with a biotin-conjugated anti-rat NTRK2 antibody serving as the detection antibody. Standards, test samples, and the biotinylated detection antibody are added to the wells subsequently and then cleared of unbound material with wash buffer. HRP-streptavidin is introduced, and the remaining unbound conjugates are removed by a washing step. TMB substrates are used to visualize the HRP enzymatic reaction, resulting in a blue product that turns yellow upon the addition of an acidic stop solution. The intensity of the yellow signal is directly proportional to the concentration of rat NTRK2 captured in the plate. The O.D. absorbance is recorded at 450 nm in a microplate reader to facilitate the calculation of the rat NTRK2 concentration.
Background:
NTRK2, also known as TrkB, is a high-affinity receptor for brain-derived neurotrophic factor (BDNF). The BDNF/TrkB signaling pathway is crucial for neuronal survival, differentiation, and plasticity in the central nervous system (CNS). The link between NTRK2 and obesity research primarily manifests in the CNS regulation of appetite: BDNF serves as a key anorexigenic (appetite-suppressing) signal. Defects in NTRK2 expression and function in the hypothalamus weaken BDNF signaling, leading to overeating, reduced energy expenditure, and severe obesity. Consequently, NTRK2 serves as a key signaling integration point in central obesity regulation pathways (acting synergistically with the MC pathway, leptin signaling, etc.).
