Description: Rat acetyl-CoA carboxylase 1 (ACACA) ELISA kit (15.625-1000 pg/mL) is a ready-to-use ELISA kit for analyzing rat ACACA levels with a detection range of 15.625-1000 pg/mL and a sensitivity of 9.375 pg/mL.
Assay Principle: The kit is based on the sandwich ELISA technique. Anti-rat ACACA antibody is pre-coated on a 96-well plate. Biotin-conjugated anti-rat ACACA antibody is used as the detection antibody. Standards, test samples, and biotin-conjugated detection antibody are subsequently added to the wells and washed with buffer. HRP-streptavidin is added, and the unbound conjugate is washed away with washing solution. TMB substrate is used to observe the HRP enzyme reaction. TMB produces a blue product catalyzed by HRP, which turns yellow upon addition of an acidic termination solution. The density of the yellow color is proportional to the amount of rat ACACA sample captured in the plate. The concentration of rat ACACA is calculated by reading the absorbance at 450 nm with a microplate reader.
Background: ACACA (also known as ACC1) is the first rate-limiting enzyme in fatty acid synthesis and is primarily responsible for converting cytoplasmic acetyl-CoA to malonyl-CoA. It plays a central role in fatty acid synthesis and is a promising therapeutic target for the treatment of obesity, cancer, metabolic diseases, and other disorders.
